The French forward scored twice in the first half to cancel out Lionel Messi's opener, before completing his hat-trick in the second half to seal a famous victory for PSG.
The result gives PSG a 5-2 aggregate lead heading into the second leg of their last-16 tie, and they will be confident of reaching the quarter-finals for the first time since 2016.
Barcelona, meanwhile, will be left to rue a number of missed chances, as well as the sending off of Clement Lenglet in the first half.
The match started brightly, with both teams looking to attack from the off.
Barcelona took the lead in the 27th minute when Messi curled a free-kick into the top corner.
However, PSG hit back almost immediately, with Mbappe equalising just three minutes later.
The French forward then put PSG ahead in the 32nd minute, finishing off a fine move involving Neymar and Angel Di Maria.
Barcelona were dealt a further blow in the 37th minute when Lenglet was sent off for a second yellow card.
PSG made the most of their numerical advantage in the second half, with Mbappe completing his hat-trick in the 65th minute.
The French forward then turned provider for Mauro Icardi, who added a fourth goal for PSG in the 85th minute.
The result was a famous victory for PSG, and they will now be confident of reaching the quarter-finals of the Champions League for the first time since 2016.
